What Lipinski Rule of Five Means
Lipinski's Rule of Five is a quick oral drug-likeness guideline. A compound is commonly considered more rule-of-five-like when molecular weight is at or below 500, LogP is at or below 5, hydrogen-bond donors are at or below 5, and hydrogen-bond acceptors are at or below 10.
- Rules are screening guidelines, not clinical or biological guarantees.
- Many useful drugs and natural products intentionally violate one or more rules.
