A thin layer of onion epidermis on a slide with a drop of iodine, a cover slip lowered at an angle, then viewed on a compound microscope. GCSE / high school; A-level / AP / IB Biology.

What the template shows

  • Mount and stain onion epidermis cells and view them under a light microscope
  • Light microscope
  • Start on the lowest-power objective; focus with the coarse knob by moving the stage away from the lens, then fine focus
  • Clip the slide over the light. Magnification = eyepiece × objective
  • Peel one cell layer of epidermis from the inside of an onion scale leaf
  • Iodine solution
  • Stains starch and nuclei so the cells show up
  • Slide and cover slip
  • Spread the layer flat in a drop of iodine. Lower the cover slip at an angle with a mounted needle so no air bubbles are trapped; blot off excess
  • Reaction setup
