Enzyme activity: amylase and starch template

Premium template. Preview and edit it free in MolDraw; downloading and exporting it is part of MolDraw Pro.
Test tubes of starch solution with a pH buffer and of amylase warming in a water bath with a thermometer, sampled every 30 s onto a spotting tile of iodine; stopwatch. Variant: catalase on hydrogen peroxide, O₂ measured with a gas syringe. GCSE / high school; A-level / AP / IB Biology.
What the template shows
- Time how long amylase takes to digest starch at different pH values (or temperatures)
- Starch + buffer
- Amylase solution
- Warm both tubes to the bath temperature, then mix and start the stopwatch
- Keep the water bath at about 35 °C (or vary it to test temperature)
- Every 30 s, one drop of mixture into a fresh well; rinse between samples
- Spotting tile
- A drop of iodine solution in each well. Blue-black = starch left; stays orange-brown once digestion is complete. Rate = 1/t. Catalase variant: H₂O₂ on yeast or potato, O₂ into a gas syringe
